Melanotan II
MT-II
Melanotan II activates several melanocortin receptors rather than a single selective target: MC1R, MC3R, MC4R and MC5R. MC1R drives melanin production and tanning; MC4R contributes to erections and appetite reduction. Lack of MC2R selectivity is associated with adrenal adverse effects.

What is Melanotan II?
Melanotan II, also called MT-II, is a synthetic α-MSH analogue investigated for pigmentation, erectile responses and appetite reduction. Serious concerns include severe nausea, uncontrolled changes in melanocytes and moles, high blood pressure and priapism. FDA warnings have been issued repeatedly. It has no FDA approval, and 503A pharmacies cannot compound it. Exercise extreme caution.
Research schedules name a low starting amount and a titrated standard amount without numeric doses. Frequency depends on the protocol, and the route varies.
Responses differ between individuals. Begin with the lowest dose and observe your response carefully. Speak with your clinician before use and report unexpected symptoms. Human adverse effects are not fully characterized for every compound.
Mechanism of action
Melanotan II activates several melanocortin receptors rather than a single selective target: MC1R, MC3R, MC4R and MC5R. MC1R drives melanin production and tanning; MC4R contributes to erections and appetite reduction. Lack of MC2R selectivity is associated with adrenal adverse effects.

How to reconstitute and inject
Diluent: Use bacteriostatic water; avoid saline because precipitation may occur.
- Let the refrigerated vial stand at room temperature for 15–20 minutes before preparation.
- Disinfect the stopper using an alcohol swab, then wait until it has dried fully in the air.
- Measure the calculated quantity of bacteriostatic water with a sterile syringe. Use only this diluent; avoid saline because precipitation may occur.
- Add the water gradually against the inner vial wall. Keep the stream away from the powder itself.
- Rotate the vial with a gentle swirling motion. Do not shake forcefully, because this can degrade the contents.
- Wait 2–3 minutes for the powder to dissolve completely, and check that the liquid is transparent and has no color.
- Use an insulin syringe to withdraw the calculated amount for injection beneath the skin.
- Inject in the thigh, abdomen or upper arm. Change the injection location each time to help prevent lipodystrophy.
- Refrigerate the mixed solution at 2–8°C. Use it within a maximum of 28 days after preparation.
What to expect
- Week 1–2
- During the first 1–2 weeks, early effects may accompany an adjustment period. Watch for adverse reactions.
- Week 2–4
- By weeks 2–4, the main effects may start becoming apparent. Make dose changes only according to your clinician’s guidance.
- Week 4–8
- During weeks 4–8, the principal effects are expected to be established. Review your progress with your clinician.
- Week 8+
- From week 8 onward, review the results and decide with your clinician whether to continue, cycle or change the dose.
Side effects and safety
- Mild pain, bruising or redness around the injection location
- Possible allergic reaction; stop use if hives or a rash develops

Frequently asked questions
What is Melanotan II?
MT-II is investigated for tanning, erectile responses and reduced appetite. Severe nausea, high blood pressure, priapism and uncontrolled mole changes are major concerns. FDA warnings and the prohibition on 503A compounding accompany its unapproved status.
Which benefits are being investigated?
Research and clinical observations indicate effects on sexual responses. Clinical observations suggest improvements in measures of desire and arousal. Its activity involves melanocortin receptors within the central nervous system. Evidence strength differs by outcome; these areas of investigation do not guarantee benefit for an individual.
Which dosing schedules are discussed?
An injectable range of 0.5–1 mg appears in research discussions, with an approximate half-life of 33 minutes. This use is not recommended and warrants extreme caution.
What safety issues matter?
Adverse effects vary with dose, administration route and individual response. Injection reactions may occur. Discuss unusual symptoms and any proposed combination of medications with your clinician before continuing.
